艾烟对SAMP8小鼠脑乙酰胆碱转移酶和胆碱酯酶活性的影响

摘    要:目的:观察艾烟对快速老化模型小鼠SAMP8脑乙酰胆碱转移酶(ChAT)和胆碱酯酶(AChE)活性的影响。

方法:选用6月龄SAMP8小鼠40只,随机分为3个艾烟干预组(低浓度组、中浓度组、高浓度组)及模型组,每组各10只。另以10只同月龄SAMR1小鼠作为正常组。3个艾烟干预组小鼠每天采用相应浓度的艾烟干预30分钟,一周6次,连续干预4周。模型组和正常组小鼠不用艾烟干预。比色法检测大脑ChAT和AChE的活性。结果:与正常组相比,模型组小鼠脑ChAT活性显著降低(p<0.01),AChE活性显著升高(p<0.01)。与模型组相比,低浓度组和中浓度组ChAT活性显著升高(p<0.05),但AChE活性无显著差异;高浓度组ChAT和AChE活性均与模型组无显著差异。3个艾烟干预组之间ChAT和AChE活性差异无统计学意义。结论:艾烟可通过对乙酰胆碱代谢的调节起到一定的延缓衰老的作用,其干预浓度以低浓度和中浓度为宜。

Abstract: Objective: To study the effect of moxa smoke on brain ChAT and AChE activity of SAMP8 mouse. Methods: 40 SAMP8 mouse were randomly divided into 3 intervention groups (i.e. low concentration group, middle concentration group, high concentration group) and one model group with 10 cases in each group. 10 age-matched SAMR1 mouse were used as normal group. Mice in 3 intervention groups were exposed to moxa smoke of different concentration for 30 minutes each time, and fumigation was done 6 times a week for 4 weeks. Mice in model group and normal group were not exposed to moxa smoke. The content of brain ChAT and AChE activity were tested by colorimetric method. Results: Compared with normal group, model group showed a marked decrease on activity of ChAT (p<0.01) and a remarkable increase on activity of AChE (p<0.01). Compared with model group, Activity of ChAT was significantly higher in low and middle concentration group than in model group. There were no difference between high concentration group and model group on activities of ChAT and AChE. 3 intervention groups showed no significant difference on activities of ChAT and AChE either. Conclusion: moxa smoke can exert anti-aging effect through metabolic adjustment of Ach, and its optimum condition should be low concentration and middle concentration.
